In this work, a simple method hydrazide-capturing technique followed by MALDI MS and MS/MS analysis for characterization of major pollen allergens of Olea europaea pollen was exploited. The strategy developed has different stages: devising of sample preparation protocol to sequester glycoproteins from a crude extract, and allowing the identification of antigenic peptides, glycosylation sites and modified glycans by MALDI MS and MS/MS.
